Anti-inflammatory Activity of Agaricus blazei Extract in Lipopolysaccharide-stimulated RAW264.7 Cells

Abstract

In this study, antioxidation and anti-inflammatory activity Agaricus Brazei (AC) extract were examined. Antioxidation activity was increased as the concentration of AC extract increased (Concentration-dependant manner). The contents of phenolic compounds and flavonoids were 111.3019 mg/g and 15.473 mg/g, respectively. In anti-inflammatory effect test using AC extract, no toxicity was shown. NO production was inhibited in range of 50 to 100 ¥ìg/mL of AC extract as concentration dependant manner. PGE2 and Proinflammatory cytokines IL-6 production was decreased as concentration dependant manner. However, TNF-¥á production was not changed compare to control. iNOS and COX-2 expression, representative factors of signal transduction of inflammatory effects, were decreased as dose-dependant manner. These result indicates that AC extract is involved in regulatory pathway of iNOS, which is related to NO, and COX-2, which is related to PGE2. As a result, AC extract is useful material which has anti-oxidant effect and anti-inflammatory effect with no toxicity.
